摘要 目的 观察鱼藤酮对大鼠肾上腺嗜铬细胞瘤细胞 (pheochromocytoma ,PC12细胞 )线粒体膜电位及细胞周期的影响 ,探讨鱼藤酮对多巴胺神经元的损伤机制。方法 用流式细胞仪检测PC12细胞线粒体膜电位及细胞周期。结果 5 0 μmol/L鱼藤酮作用于PC12细胞 12h及 2 4h后 ,G0 /G1期及S期细胞所占比例逐渐下降 (P <0 0 1) ,而G2 /M期细胞比例逐渐升高 (P <0 0 1)。单纯鱼藤酮中毒 12h线粒体膜电位即降低。结论 大剂量 (5 0 μmol/L)鱼藤酮引起线粒体膜电位降低及细胞周期改变 ,可能导致PC12细胞生长障碍。 Objective To observe the effects of rotenone on the mitochondrial membrane potential and cell cycles in pheochromocytoma (PC12) cells and to explore the injury mechanism of rotenone on dopaminergic neuron. Methods The mitochondrial membrane potential and cell cycles were determined by flow cytometry. Results After treatment with 5.0 μmol/L rotenone for 12 and 24 h, the percentage of G 0/G 1 phase and S phase in cultured PC12 cells decreased gradually (P<0.01), but at the same time, the percentage of G 2/M phase increased. The mitochondrial membrane potential decreased markedly after administration of rotenone alone for 12 h. Conclusion Rotenone at the dose of 5.0 μmol/L can induce the decrease of mitochondrial membrane potential and the changes of cell cycles, resulting in the inhibition of PC12 cell growth.
