缺氧诱导人成骨细胞VEGF的表达

摘要 目的 探讨缺氧对人成骨细胞VEGF表达的影响。方法 本实验将手术中取得的人髂骨块 ,使用酶消化法进行培养。当细胞传至第二代时 ,建立人成骨细胞缺氧模型 ,采用免疫组织化学方法、计算机图像分析仪检测人成骨细胞VEGF的表达。结果 在培养的前 3d ,缺氧组VEGF免疫组织化学染色阳性细胞的平均灰度低于对照组 ,两组比较差异有显著性 (P <0 0l)。结论 缺氧 (1~ 3d)诱导人成骨细胞VEGF的高表达。 Objective This study is to investigate the expression of VEGF after exposure of human osteoblasts to anoxic stimuli . Methods The osteoblasts obtained from the human iliac bone during operation were cultured by the method of enzyme digestion in vitro.After establishment the hypoxic model in osteoblasts of the second generation. Detect the expression of VEGF of human osteoblasts by immunohistochemistry study and image analyzer. Results When the cells were cultured for the first three days , the average grayscale of positive cells of anoxic group are lower than normal group .The difference of two groups was significant(P<0.01). Conclusions Hypoxia(1~3 days)can induce the high expression of VEGF of human osteoblasts.
