摘要
目的探讨全身炎性反应综合征(SIRS)大鼠心肌刺激性G蛋白(Gs)含量、GsmRNA表达的变化及地塞米松对Gs变化的影响。方法实验组大鼠给予大肠杆菌脂多糖(LPS)20mg/kg腹腔注射,制造SIRS大鼠模型。地塞米松(DXM)组在同样给予LPS后10min给地塞米松20mg/kg腹腔注射。免疫印迹法测定心肌Gs含量,RTPCR半定量法检测GsmRNA的表达。结果与对照组相比,LPS3h组、LPS6h组大鼠心肌Gs的相对含量下降,尤以6h组下降明显(P<0.01)。DXM组与LPS3h组比较,Gs的相对含量略有回升,但无统计学意义。与对照组相比,LPS3h组、LPS6h组GsmRNA的表达值均有显著性差异(P均<0.01),尤以6h组下降明显。DXM组与LPS3h组相比,较前者略有好转,但无统计学意义(P>0.05)。结论SIRS大鼠心肌细胞信号转导系统中的Gs发生异常,此种异常改变是在基因水平上发生的,并且参与了心功能障碍的发生、发展。20mg/kg剂量的地塞米松对Gs的变化没有明显的影响。
Objective: To explore the alterations of myocardial stimulatory G protein(Gs) and its mRNA expression in systemic inflamatory response syndrome (SIRS) rats and the effect of dexamethason (DXM). Methods: The SIRS model of rats was made by lipopolysaccharide (LPS) injection, and DXM treatment group received DXM 20 mg/kg intraperitoneally. Tissue samples from heart were collected 3 and 6 hours after the injection of LPS. Content of Gs was measured by using western blot analysis. Gs mRNA expression was detected with the method of reverse transcription polymerase chain reaction (RT-PCR). Results: Compared with the control group, the content of Gs in LPS 3 h group and LPS 6 h group were significantly decreased (all P<0.01). The change was more obvious in LPS 6 h group. It was unchanged in the DXM group (P>0.05). In LPS 3 h group and LPS 6 h group, the expression of Gs mRNA was decreased significantly (all P<0.01) when compared with the control group. The change was more obvious in LPS 6 h group. It was unchanged in the DXM group (P>0.05). Conclusion: With the development of the SIRS, the cardiac functions of rats were injured progressively. The content of Gs and its mRNA expression were decreased and these changes may contribute to the occurrence and development of the cardiac dysfunction of the SIRS rats. DXM of 20 mg/kg has no effect to the alterations of the myocardial Gs and its expression.
