HCV HVR1准种在不同基因型丙型肝炎患者中的分布

Distribution of HCV HVR1 quasispecies in patients infected with HCV of different genotypes

目的通过研究丙型肝炎病毒高变区1(HCVHVR1)准种复杂性与基因型之间的关系,探讨不同基因型HCV致病性差异的机制。方法采用C区型特异性引物PCR法进行HCV基因分型,采用单链构象多态性聚合酶链反应法(SSCP法)检测HCVHVR1准种。结果68例丙型肝炎患者中,Ⅱ型49例(72%),Ⅲ型13例(19%),Ⅱ、Ⅲ混合型6例(9%)。Ⅱ型和Ⅱ、Ⅲ混合型HCV感染患者的HCVHVR1准种复杂性(SSCP条带数)明显高于Ⅲ型感染的患者,且与疾病严重程度关系密切。结论HCVHVR1准种复杂性的差异可能是导致不同基因型HCV致病性差异的因素之一。

Objective: The relationship between HCV HVR1 quasispecies complexity and HCV genotypes was investigated in order to study the difference of pathogenicity of HCV of different genotype.Methods:The differentiation of HCV genotype was detected by using polymerase chain reaction with core region type-specific primers. HVR1 quasispecies were examined with the method of polymerase chain reaction-mediated single-strand conformation polymorphism (SSCP). Results:Forty-nine(72%) of 68 patients were infected with HCV of genotype Ⅱ, 13(19%) with HCV of genotype Ⅲ, 6(9%) with coinfection of genotype. The complexity of HCV HVR1 quasispecies(number of SSCP bands)was higher in patiants with genotype Ⅱ and coinfection than that in those with genotype Ⅲ, and the complexity was related with the progress of disease.Conclusion:The different distribution of HCV HVR1 quasispecies in different genotypes may be one of factors of different pathogenicity in HCV.
