乳杆菌代谢物对白色念珠菌影响的体外试验研究 被引量:4

摘要 目的 从阴道微生态平衡角度出发 ,观察乳杆菌 (DM9811)代谢物对白色念珠菌的抑杀效果 ,并行组份分析 ,探索CV(candidalvaginitis,CV)的生态疗法。方法 取CV患者阴道分泌物 30例 ,分离培养 ,鉴定 ,保留白色念珠菌菌株。 1 以提取液 (以下简称提取液 )Ⅰ和Ⅱ分别代替蒸馏水配制培养基SⅠ 和SⅡ ,将保留的白色念珠菌株传代 5次后 ,配制菌液 ,接种于S、SⅠ 和SⅡ 中 ,培养后活菌计数。 2 对提取液Ⅰ和Ⅱ倍比稀释 ,并代替蒸馏水配制多个培养基SⅠ 和SⅡ ,测定SⅠ 和SⅡ 中没有菌落生长的最低有效浓度 (MIC)。 3 气相色谱测定提取液中挥发酸类成分并定量。 4 选择提取液最低浓度加 1的方法配制培养基 ,比较提取液和乙酸水溶液对白色念珠菌的抑制效果。结果  1 白色念珠菌生长 2 4h后计活菌数 ,将SⅠ 与S ,SⅡ 与S中活菌数相比较 ,非参数配对T检验P <0 0 1。 2 提取液Ⅰ最低有效浓度为 1∶4 ,提取液Ⅱ最低有效浓度为 1∶2。 3 气相色谱测定提取液Ⅱ中挥发酸类主要成分为乙酸 ,浓度为 0 2 35 9S/10 0ml。 4 提取液和乙酸对白色念珠菌的生长都有抑制作用 ,但提取液的抑制作用明显强于乙酸。结论 乳酸杆菌DM9811代谢物对白色念珠菌有明显的抑菌和杀菌作用 ,可作为一种新型的生态制剂进行开发 ,用于治疗? Objective To study the effects of inhibition and disinfection of Candida albican by the metabolite of Lactobacillus DM9811 in vitro and analysis components of metaboilte. Methods To scrape their vaginal secretion and the isolate,culture,identify and preserve 30 strains of Candida albicans from the secretion.1.To dispense sabouraud's culture liquid named 'S',and to dispense sabourud's culture liquid in which the extract I and II of the metabolite of lactobacillus respectively replace distilled water named 'S_I' and 'S_ II '.The preserved Canduda albicans were 50 μl of the solution of Candida albicans into 5 ml of S_I,all and S,and cultured in incubator at 37 ℃ for 24 hours,and to count the number of bacteria.2.S_I and S_ II were diluted doubly every time and determined the minimum inhibitory concentration(MC) where no bacteria were found.3. To detect volatile acid of extract II of the metabolite of lactobacillus and quantitative analysis by gas chromatography.4.The detected volatile acid was dispensed to liquide'Y' by the concetration in the extract II. To dispense sabourand's culture liquid 'S_Y' in which liquide Y replaces distilled water.S_ II and S_Y were diluted by the concentration of minimum inhibitory concentration plused one named S_ II * and S_Y *.To inoculate 50 μl of 10 8CFU/ml of Candida albicans into 5 ml of S,S_ II * and S_Y *,cultured in incubator at 37 ℃,and to count the number of bacteria every two hours,total 24 hours amounting to 12 points.To draw the growth curves of Candida albicans in S,S_ II * and S_Y *. Results 1.The number of Candida albicans cultured in S,S_ I * and S_ II for 24 hours were obviously different with nonparametric T-test between S_I and S,S_ II and S(P<0.01).2.The minimum inhibitory concentration(MIC) of the extract I was 1∶4,the extract Ⅱ was 1∶2.3. The major component of the extract II of the metabolite of lactobacillus was acetic acid detected by gas chromatography,its concentration was 0.235 9 g/100 ml.4.The growth curves of Candida albicans in S,S_ II * and S_Y * indicate the the growth of Candida albicans was inhibited by S_ II * and S_Y *,and the inhibition obvious by S_ II * than S_Y *. Conclusions The metabolite of lactobacillus DM9811 had obvious effects of inhibition and disinfection on Candida albicans and provide a new ecological agents to treat candida vaginitis.
