摘要
本文应用抗普通型急性淋巴细胞性白血病细胞抗原的单克隆抗体(McAb79)与蓖麻毒素(Ricin)连结制备了免疫毒素(IT),并用IT特异地杀伤白血病细胞(Nalm—1)。~3H—亮氨酸掺入试验说明McAb79:Ricin结合物能够明显地抑制白血病细胞的蛋白质合成,从而表现了特异的细胞毒作用。集落形成抑制试验提示McAb79:Ricin结合物能显著地抑制白血病细胞的集落形成,而无关抗体N34:Ricin结合物仅轻度抑制白血病细胞集落形成(P<0.01),表明McAb79:Ricin结合物能够在体外有效的杀灭白血病细胞(Nalm—1)。
The immunotoxins that specially kill leukemic cells(Nalm-1 ) wereprepared by conjugating ricin with monoclonal antibody 79 directed to common acute lympho cytic leukemic cell antigen. 3 H-L-Leucin incorporation test showed that McAb 79: Ricin conjugate could apparently inhibit protein synthesis of leukemic cells and express specific cy-totoxic effects. The inhibition test of clonogenic growth suggested that McAb 79: Ricin con jugate could markedly inhibit clonogenic growth of leukemic cells, while the control conjugate showed only slight inhibition to clonogenic growth of leukemic cells. The difference was statistically significant (P<0.01).These results indicated that leukemic cell (Nalm-1) may be effectively killed by McAb 79: Ricin conjugate in vitro.
