低温冷冻保存角膜的超微结构观察 被引量:4

摘要 应用扫描电镜和透射电镜,对经不同低温冷冻方法保存及不同保存时间的14例角膜进行了超微结构观察。结果发现,扫描电镜下,除1例为甘油法保存473天者,其角膜内皮细胞大部分被破坏外,其它标本的内皮改变程度与保存方法无明显关系;透射电镜下,观察到角膜内皮细胞的有10例,内皮细胞质膜除1例外,其它均完整,内皮细胞、基质细胞及上皮均有线粒体肿胀、内质网池扩张和胞质内空泡形成,唯有内皮细胞的改变程度较后二者更为明显。 Ultrastructural observation was carried out on 14 cases ofcornea preserved in different cryoprotectants and for different dur-ations by screening and infiltrating ultramicroscopes.The resultsshowed that under the screening microscope nearly all the endo-thelial cells of cornea were destroyed except in one case cryopre-served in glycerine for 473 days and that there seemed to be no appar-ent difference or relationship amon gdifferent methods of cryopreser-ration.Under the infiltrating ultramicroscope,the endothelial cellsof cornea were observed in 10 cas.esThe cytomembranes of theendothelial cells were all intact with the exception of one case.Themitochondria were swollen and there were dilatation and cysticformation of the endoreticulum in the endothelial stromal andepithelial cells.Only the changes in endothelial cells were more pro-minent than in other types of cells.
