摘要
研究了荧光素合成过程中浓硫酸的用量及其粗制品提纯方法的改良,并对其纯品进行了动物及临床试验。结果表明,浓硫酸的最佳用量为6ml,反应速度可加快1/3;用氢氧化钠代替乙醇溶解,成本为乙醇的1/10;用醋酸代替乙醇和盐酸使荧光素析出,可使其产率提高到85%;临床试验69例,荧光显影清晰,荧光持续时间为15~60min,无不良反应。提示该产品优于目前国内同类产品。
Fluorescein was synthesized by treatment of resorcinol with O-phthalic anhydr-ide in the presence of concentrated sulfuric acid.The mass was then disintegra-ted and boiled in water to remove soluble impurities.Fluorescein in distilled waterwas adjusted to pH 8.0~8.5 with sodium hydroxide solution and filtered,and thefiltrate,treated with 1% active charcoal,was filtered again and mixed withdistilled water to form a 105 soltion of sodiun flaorescein,Then it was filledinto 5ml ampuls and sterilized at temperature of 100℃ for 30 minutes to producesodium fluorescein for i. v. injection.This injection gave a clear-cut developmentand the fluoresce lasted for 15~60mins.The side-ecffet is much less than thoseof other oroducts nresently available in this country.
