摘要
通过检测MKN28人胃癌细胞株~3H-胸腺嘧啶核苷掺人情况,在体外实验研究加温和抗癌药物顺氨氯铂、5-氟脲嘧啶之间的协同作用。结果表明(1)顺氨氯铂的细胞杀伤作用具有温度-药物浓度依赖性;(2)5-氟脲嘧啶合并加温未见有显著协同作用,反见削弱其抗肿瘤效应;(3)单纯加温与合并应用中等剂量的顺氨氯铂或5-氟脲嘧啶的比较中发现,效果最佳的为加温加顺氨氯铂,其次是单纯加温;(4)加温加顺氨氯铂、5-氟脲嘧啶组合治疗效果最佳。为临床应用治疗胃癌的腹腔广泛转移提供了理论依据。
The effects of combined application of hyperthermia, CDDP and 5-FU on MKN28 human gastric cancer cell line in vitro were investigated. Results showed: (1) CDDP caused a substantial decrease in the incorporation of ~3H-TdR in a concentration-and temperature-dependent mannex. (2) No synergy in the cytotoxic effect was noted when 5-FU was used in combination with hyperthermia. (3) Effects of hyperthermia in combination with CDDP were better than those of either hyperthermia alone or hypcrthermia combined with 5-FU. (4) Combined application of hyperthermia, CDDP and 5-FU showed a maximal effect on cell's incorporation of ~3H-TdR vs that with only one or two of these modelities. These results supported the impression that hyperthermia combined with anti-cancer drugs had synergistic effect against gastric cancer with disseminated peritoneal metastases.
