Effects of Vitamin C and Trolox on Adriamycin-induced Cardiotoxicity in Cultured Rat Myocardial Cells

摘要 在原代培养的大鼠心肌细胞受阿霉素损伤的模型上,观察到合适浓度的维生素C(Vit C)能显著减少受损心肌细胞乳酸脱氢酶释放、减少细胞内丙二醛的形成,并改善细胞超微结构的变化,显示其具有抑制脂质过氧化,保护心肌细胞的作用.该作用与Vit c浓度密切相关,高浓度时反而加重细胞损伤.维生素E的水溶性类似物Trelox则没有上述作用。 Primary cultures of rat myocardial cells were used to investigate the effects of ascorbic acid and trolox on the cardiotoxicity of adriarnycin (ADM). The 3-day cultured myocardial cells with ADM (1.8μmol/L) were treated with different doses of ascorbic acid (0.056, 0.17, 0.56, 0.17mmol/L) and trolox (0.05,0.1,0.2mmol/L). After being incubated for 4h, the lactic dehyrogenase(LDH) activity in the media and the content of malondialdehyde (MDA) in myocardial cells were assayed. The ultrastructural changes of the cells were also scrutinized. It was shown that the LDH activity in the media with the addition of ascorbic acid (0.56mmol/L) was significantly lowered vs that treated with APM aione, and that the MDA contant in the cells was reduced. THe ultrastructural impairments in the myocardial cells were much improved after the addition of ascorbic acid. The effects of ascorbic acid were dose-dependent, while overdosage could induce peroxidation. Trolox had no significant effect on ADM-induced LDH release and MDA rise, nor did it affect the ultrastructural changes of the myocardial cells.
