摘要
目的 探讨川芎嗪对培养大鼠星形胶质细胞在氧糖剥夺损伤中的保护作用及可能机制。方法 利用氧糖剥夺 (OGD)建立星形胶质细胞缺血再灌损伤模型后 ,分别测定细胞培养上清中一氧化氮 (NO)、乳酸脱氢酶 (LDH)浓度。四甲基偶氮唑盐 (MTT)代谢率测定星形胶质细胞活力。免疫组化检测核转录因子 -κB(NF -κB)活化程度。结果 川芎嗪干预组与模型组比较 ,NO分泌量、LDH漏出、细胞活力下降均明显减少 (P <0 0 1) ;P6 5胞核移位明显减少 (P <0 0 1)。结论 川芎嗪对培养大鼠星形胶质细胞在氧糖剥夺损伤中具有保护作用 ,其作用机制之一可能是通过抑制NF -κB活化 ,减少NO分泌来实现的。
Objective To investigate the protective effect and its possible mechanism of Tetramethylphyrazine on cultured rat astrocytes damaged by oxygen glucose deprivation. Methods We used OGD to malce the model of ischemical reperfusion injury in rat astrocytes. NO and LDH concentrations in the supernatant of cultured cells were measured separately. Vigors of astrocytes were determined by MTT metabolic rate. Immunohistochemistry assay was performed to test NF-κB activation levels. Results Compared with the model group, NO formation, LDH leakage and descent of cellular vigor were all significantly decreased in Tetramethylphyrazine intervention group (P<0.01); P65 transposition of nucleus was also reduced (P<0.01).Conclusion Tetramethylphyrazine had protective effects on astrocytes damaged by OGD, one of the mechanisms might be related with its inhibition of NF-κB activation to decrease secretion of NO.
