摘要
目的:对1998年本室建立的人肝癌细胞系HCC鄄9810进行细胞遗传学分析。方法:采用组织块培养法,培养液为含20%的胎牛血清的RPMI鄄1640,用扫描电镜和透射电镜观察细胞形态熏放射免疫方法检测肿瘤标志物,并用改良的热变性姬姆萨R显代穴RHG雪方法与G显带相结合进行细胞遗传学检测。结果:该细胞系经异种接种能形成移植性肿瘤,染色体为非整倍体,有4q鄄熏11q鄄熏13q+等。结论:检出的这些畸变可能与肝癌的发生、发展有关。
Objective: To explore the relation between chromosomal aberration and carcinogenesis of the liver by molecular cytogenetic analysis of the hepatocellular carcinoma cell line. Methods: Small tissue blocks taken from resected specimens of a patient with hepatic cancer were cultured in medium RPMR-1640 added with 20% fetal bovine serum, scanning electron microscopic and transmission electron microscopic used to observe the cells, and tumor markers were detected by radioimmunoassay. Molecular cytogenetic studies were performed on the cell line using chromosome R-banding and G-banding. Results: The cell line was aneuploid. 4q-,11q-,13q+ were observed in 60%~90% of the cells. Conclusion: The presene of the reported chromosomal aberration may be related to the carcinogenesis and cancer development of the liver.
