摘要
用半制备型反相高效液相色谱(RPLC)对箭叶淫羊藿提取液中的 5种主要黄酮类化合物进行了分离制备,优化了分离条件。每次进提取液 35mL, 经一步RPLC便可得到纯度均大于 95%的化合物Ⅰ、Ⅱ、Ⅲ和Ⅴ,经过 10次制备,共制得 93mgⅠ、71mgⅡ、296mgⅢ和 487mgⅤ。而经一步纯化得到的化合物Ⅳ的纯度较低,再经第二步RPLC纯化后可使其纯度大于 95%,最后得到 64mgⅣ。经波谱分析鉴定所得的 5种化合物分别为hexandrasideF、朝藿定A、朝藿定B、朝藿定C及淫羊藿甙,均为 8 异戊烯基黄酮类化合物,其中hexandrasideF是首次从淫羊藿属植物中分离得到的。
Five flavonol glycosides in the extract of Epimedium sagittatum were isolated and prepared by semi-preparative reversed phase liquid chromatography (RPLC) and the chromatographic conditions were optimized. When 35 mL of extract were injected into the reversed phase liquid chromatogrpahy (RPLC) column, the purities of compound I (hexandraside F), II (epimedin A), III (epimedin B) and V (icariin) were all found to be more than 95% only by one run with RPLC. After ten RPLC runs, 93 mg of I, 71 mg of 11, 296 mg of M and 487 mg of V were obtained, respectively. However, the purity of compound IV (epimedin C) can be obtained to be more than 95% by re-separation by RPLC and 64 mg of compound IV was obtained finally by ten time runs. Their structures were identified and confirmed to be the foregoing five compounds of flavonol glycosides. In addition, hexandraside F was firstly isolated from the genus Epimedium.
