中药金叶败毒制剂对人巨细胞病毒感染细胞及细胞周期调控抑制基因WAF1/CIP1表达的研究 被引量:1

摘要 目的 通过研究中药金叶败毒制剂对人巨细胞病毒 (HCMV)感染细胞及细胞周期调控抑制基因WAF1/CIP1蛋白丰度变化的影响 ,探讨中药金叶败毒制剂体外对HCMV感染的阻断和治疗效应及其机制。方法  2 0 0 2年 11月至 2 0 0 4年 4月华中科技大学同济医学院附属同济医院采用蛋白免疫印迹 (Westernblot)方法检测HCMV感染后 2 4h、4 8h、72h、96h细胞中p2 1WAF1/CIP1丰度 ,检测中药金叶败毒制剂干预后p2 1WAF1/CIP1丰度的变化。结果 病毒感染后 2 4h、4 8h、72h、96h细胞中 p2 1WAF1/CIP1丰度呈下降趋势 ,而中药金叶败毒制剂可部分阻止这种趋势。结论 中药金叶败毒制剂部分上调HCMV感染引起的p2 1WAF1/CIP1丰度下降 ,可能是中药金叶败毒制剂抗HCMV效应的作用机制之一。 Objective The effects of Chinese traditional herb Jinyebaidu on the abundance of control inhibiting gene p21WAF1/CIP1 protein variation of concentration in the cell cycle of infected HCMV cells,moreover,the block,therapy effects and mechanisms of Chinese traditional herb Jinyebaidu against virus infection cell in vitro were investigated.Methods The abundance of p21WAF1/CIP1 protein of the infected HCMV cell after 24h,48h,72h,96h were measured by western blot from November 2002 to April 2004 in the Department of OBGY in Tongji Hospital Affiliated to Tongji Medical College,Huazhong Science and Technology University,and the change of the abundence of p21WAF1/CIP1 protein interfered by Chinese traditional herb Jinyebaidu was detected.Results The abundance of p21WAF1/CIP1 protein of infected HCMV cells showed the trend of decreasing from 24h to 96h,but Chinese traditional herb Jinyebaidu could partly arrest this trend.Conclusion Chinese traditional herb Jinyebaidu can partly up regulate the abundance of p21WAF1/CIP1 protein in HCMV infected cell,therefore it may be one of the antivirus mechanisms.
