摘要
目的 建立大鼠面神经损伤实验动物模型 ,观察重组Neurturin对面神经元的营养与保护作用。方法 以切断面神经总干的方法 ,造成大鼠两侧面神经损伤模型 ,在右侧面神经损伤处给予重组Neurturin ,左侧作为对照侧 ,给予PBS ,分别于手术后 7、14、2 8和 4 2d ,取脑组织行双侧面神经核切片 ,甲苯胺兰染色 ,观察面神经元形态 ,分别计数两侧面神经元数量。结果 给予重组Neurturin的一侧大鼠面神经核大部分细胞仍保持正常形态 ,对照侧面神经核神经细胞多呈空泡样变性、坏死 ,部分正常形态已消失 ;手术后 7及 14d ,给药一侧存活的神经元数量明显比对照侧多 ,2 8和 4 2d差异无显著意义。结论 重组Neurturin对损伤的大鼠面神经核神经元细胞具有营养和保护作用。
Objective To establish a rat model of facial paralysis and observe the nutritional and protective effect of recombinant neurturin on neuron.Methods Establish a rat model of bilateral facial paralysis by cutting off main facial nerves. Treat the paralysis at right side with recombinant neurturin, and that at left side with PBS as a control. Take brain tissues 7,14,28 and 42 d after operation respectively for preparation of pathological sections of bilateral facial nuclei. Stain the sections with toluidine blue and observe the morphology of neuron. Count the neurons at two sides respectively.Results Most of facial nuclei cells treated with recombinant neurturin showed normal morphology, however, most of those treated with PBS showed vacuolus-like denaturalization or necrosis. The number of live neurons at the right side 7 and 14 d after operation were significantly larger than those at the left side. However, the numbers of live neurons at the two sides 28 and 42 d after operation showed no significant difference.Conclusion Recombinant neurturin showed nutritional and protective effect on neuron in rat model of facial paralysis.
