肿瘤抑制基因RECK在胃癌中的表达及临床意义 被引量:9

摘要 目的:检测肿瘤抑制基因RECK(reversioninducingcysteinerichproteinwithKazalmotifs)及MMP9在胃癌组织和正常胃组织中mRNA的表达水平并探讨其临床意义。方法:采用逆转录-聚合酶链反应(RTPCR),检测47例胃癌患者的肿瘤组织和正常胃组织中RECK基因及MMP9mRNA的表达。结果:47例胃癌患者的正常组织中RECKmRNA的表达量为0793±0012,肿瘤组织中的表达量为0312±0076,两者差异有统计学意义,P=000752;正常组织中MMP9mRNA的表达量为0425±0107,肿瘤组织中的表达量为0663±0098,两者表达差异有统计学意义,P=003171,胃癌组织中RECK基因mRNA的表达与MMP9mRNA的表达呈正相关,r=055,P<005;RECK的表达与肿瘤浸润深度和周围脏器侵犯密切相关,P<005;RECK表达水平高于0314的患者的2年生存率要显著高于RECK表达水平低者,P=0043。结论:胃癌组织中RECK基因的转录水平显著降低,RECK基因与MMP9存在某种共同的转录调节机制;RECK基因低表达的肿瘤具有更强的侵袭能力,其表达与胃癌患者的预后密切相关。RECK基因可能成为判断胃癌患者预后新的指标及肿瘤治疗的新的靶位点。 OBJECTIVE:To determine the mRNA levels of transformation suppression gene RECK and MMP-9 in gastric tumorous and contiguous nontumorous tissues.METHODS: mRNA levels were investigated by reverse transcription-polymerase reaction (RT-PCR) in all the tumorous and contiguous nontumorous tissues obtained from 47 patients with gastric carcinoma.RESULTS: Among the 47 gastric carcinoma, RECK mRNA level was 0.793±0.012 in nontumorous tissues , 0.314±0.076 in the tumorous, expression levels of RECK were significance lower in tumor tissue, P<0.05. Expression levels of MMP-9 were significance lower in nontumorous tissues (0.425±0.107) than in the tumorous (0.663±0.098,P<0.05). the mRNA levels of RECK and MMP-9 showed significances positive correlation, r=0.55,P<0.05. RECK expression showed significances correlation with the depth of tumor infiltration and local invasion, P<0.05. Patients who had high expression levels of RECK in tumorous tissue showed significantly better prognosis than those had lower expression, P=0.043.CONCLUSION: RECK has negative affection the invasiveness of gastric carcinoma, the results suggest the potential value of RECK as a prognostic molecular marker and possible the possible therapeutic or preventive agents for gastric carcinoma.
