β-七叶皂甙钠对大鼠胰腺缺血再灌注损伤胰腺微循环改变的影响 被引量:1

摘要 目的探讨β-七叶皂甙钠(SA)对胰腺缺血再灌注(IR)损伤后微循环改变的影响及意义。方法40只大鼠随机分为5组假手术组,IR1h,IR6h,SA+IR1h,SA+IR6h组。测定血清淀粉酶、脂肪酶含量;光镜、电镜观察胰腺组织结构改变;微循环显微镜检测胰腺微循环的血管直径、血流速度;测定胰腺组织干湿重比。结果①与IR1h组、IR6h组相比,SA+IR1h组、SA+IR6h组的血清淀粉酶和脂肪酶含量显著降低(P<0.05)。②SA+IR1h组与IR1h组相比、SA+IR6h组与IR6h组相比,组织病理学评分降低、细胞超微结构损伤明显减轻。③SA+IR1h较IR1h组,SA+IR6h组较IR6h组微静脉直径缩小(P<0.05)、血流速度减慢明显改善(P<0.05)。④IR1h组与SA+IR1h组相比,胰腺干湿重比减少无显著差异,SA+IR6h组较IR6h组干湿重比明显增加(P<0.05)。结论β-七叶皂甙钠对胰腺IR损伤有保护作用,其保护机制可能与改善微循环有关。 Objective To investigate the effects of sodium aescinate(SA)on pancreatic microcirculation in rats with pancreatic ischemia-reperfusion injury (IR). Methods Fourty male Spraque-Dawely (SD) rats were randomly divided into 5 groups: sham group, IR 1h group, IR 6h group, SA+IR 1h group and SA+IR 6h group. Plasma concentrations of amylase and lipase were measured. The histology of pancreas were studied under optic and electron miscroscopes. Pancreatic microcirculatory parameters were determined by intravital microscopy. The ratio of pancreatic dry weight (DW) to wet weight (WW) was examined to assess the water content. Results 1.Compared with those in IR 1h and IR 6h groups,the plasma concentrations of amylase and lipase in SA+IR 1h and SA+IR 6h groups were decreased respectively(P<0.05). 2.The histologic scores and ultrastructural injury in SA+IR 1h and SA+IR 6h groups were markedly attenuated, compared with those in IR 1h and IR 6h groups, respectively. 3.The venule diameters and blood flow rate in SA+IR 1h and SA+IR 6h groups were much better than those in IR 1h and IR 6h groups, respectively(P<0.05). 4.The ratio of DW to WW of pancreatic tissue showed no significant difference between IR 1h group and SA+IR 1h group,but the ratio was increased significantly in the IR 6h group after SA treatment(P<0.05). Conclusion SA can exert beneficial influence on pancreatic IR injury,and this seems to be related to the improvement of pancreatic microcirculation.
