A study on mouse embryo cryopreservation

目的探讨小鼠胚胎冷冻保存方法。方法用EG40、ES40、EFS403种冷冻液进行小鼠胚胎快速冷冻,比较解冻后胚胎的发育率、孵化率、着床率。结果EFS40冷冻液冷冻效果最好,与对照组相比无明显差异;ES40其次,EG40效果最差。结论EFS40冷冻液适合用于小鼠胚胎冷冻保存。

Objective To study the methods of mouse embryo cryopreservation.?Methods Rapid cryopreservative method was used to freeze mouse embryos with solutions EG40,ES40 and EFS40. The development rate and hatch rate in vitro and the implantation rate in vivo were determined with thawed mouse embryos to evaluate the 3 preservatives.?Results Best cryopreservative effect was offered by solution EFS40, with the parameters of the thawed embryos comparable to that of the control fresh embryos.ES40 ranked the next,while EG40 was the worst. The development rate, the hatch rate and the implantation rate of thawed mouse embryos were 87.1%,75.3% and 67.4% with EFS40,56.4%,41.7% and 43.9% with ES40, while 10%, 0 and 11.1% with EG40,respectively.?Conclusion EFS40 is the solution of choice for mouse embryo cryopreservation.
