胃癌单克隆抗体-羟基喜树碱免疫偶合物的制备及其细胞毒特征 被引量:1

PREPARATION OF HYDROXYCAMPTOTHECINE-MONOCLONAL ANTIBODY RSF_9 IMMUNOCONJUGATE AND CHARACTERIZATION OF ITS CYTOTOXICITY TO GASTRIC CANCER CELLS IN VITRO

摘要 采用共价交联法将胃癌单克隆抗体RSF_9与羟基喜树碱进行交联,交联物中每分子抗体可引入13±4.2分子的药物,而抗体活性保持85%以上。体外细胞毒试验显示交联物RSF_9-HCPT对胃癌细胞的杀伤是游离药物的4.7倍,而对非靶细胞的杀伤只有游离药物的1/11,表明交联物对胃癌靶细胞具较强的选择性杀伤作用。 Monoclonal antibody RSF9 against human gastric cancer was linked convalently to hydroxycamptothecine (HCPT) to form an immuno-conjugate (HCPT-RSF9). The linked molecular ratio of RSF9 to HCPT was 13±4.2. ELISA revealved that the specific immunoreactivity of the McAb was well preserved in the conjugate (over 85%). The cytotoxicity of HCPT-RSF9 to human gastric cancer cells (MKN-28) was determined by MTT method. The results showed that the cytotoxicity of HCPT-RSF9 to gas-trie cancer cells was 4.7 times that of free HCPT, while to non-target cells was only 1/11 of that of free HCPT, Suggesting highly selective cytotoxicity to target cells.
