摘要
根据α 半乳糖苷键物质及其结构组成,设计了两种饲用α 半乳糖苷酶活性的检测程序。结果表明,以棉子糖为底物的DNS法,因饲料中干扰因素较多,导致酶活性偏低。而以对硝基苯酚 α D 半乳糖吡喃糖苷底物的p NPG法,具有操作简便,灵敏度高,重现性好等特点。其测定条件为:反应系统pH4.0,反应温度40℃,反应时间10min,底物浓度5mmol/L。
According to the structure compositions of α-galacotoside, two methods for determination of α-galacotosidase activity in the feed were designed. The results showed that determination value of α-galacotosidase activity by using DNS method in which adopted raffinose as substrate was lower because of greater interference factors in the feed. In p -NPG method, p-nitrophenyl-α-D-galactospytanoside was used as substrate to determinate α-galacotosidase activity in the feed.The method was simple, high sensitive and good repeatable. The determination conditions of the method were as follows: pH in reaction system, 4.0; reaction temperature, 40℃; reaction time,10 min,substrate concentration,5 mmol/L.
