摘要 目的 研究 HBV-DNA阳性血清对肾小管上皮细胞表型转化的作用。方法 以体外培养的人近端肾小管上皮细胞系 (HK-2 )为对象 ,分别用健康人血清、HBV-DNA阳性血清刺激 ,用免疫组织化学检测角蛋白(CK)、波形蛋白 (vimentin)、α-平滑肌肌动蛋白 (α-SMA)和 型胶原的蛋白表达。结果 在 HBV-DNA阳性血清直接作用下 ,肾小管上皮细胞免疫组化染色显示 CK弱阳性 ,vimentin、α-SMA和 型胶原增强。结论 在HBV-DNA阳性血清直接作用下 ,体外培养的正常人近端肾小管上皮细胞系 HK-2可发生上皮向间叶的表型转化。 Objective To study the effect of HBV-DNA positive serum on phenotypic transformation of renal tubular epithelial cells. Methods The n ormal human kidn ey proximal tubular cell line (HK-2) was cultured for 3 days in the presence of the serum of healthy person or the HBV-DNA positive serum. Immuno-histochemi stry stain was used to examine the expression of cytokeratin, vimentin, α-SMA and collagen protein. Results Being cultured in HBV-DNA posit ive serum, the r enal tubular epithelial cells showed the reduction of cytokeratin, and enhanceme nt of vimentin,α-SMA and collegen by immuno-histochemistry stain. Con clusion HBV-DNA positive serum would induce phenotypic transformation of renal tubular epithelia l cells into mesanchymal cells.
