狼疮Ⅰ方预防氢化可的松对软骨细胞的损伤作用

Effect of Lupus Fang Ⅰ on Preventing Hydrocortisone Damage to Chondrocyte

目的探讨狼疮Ⅰ方预防氢化可的松对软骨细胞的损伤作用。方法取3～4周龄兔关节软骨细胞,建立软骨细胞体外培养体系,经阿新蓝、甲苯胺蓝染色鉴定后随机分成氢化可的松组、中西药组、中药组、空白对照组。分别培养12h和36h,用倒置显微镜观察软骨细胞形态变化,用MTT比色法观察软骨细胞OD值的变化。结果12h和36h氢化可松组软骨细胞严重损伤或死亡;中西药组、中药组、空白对照组软骨细胞生长良好。MTT检测结果与软骨细胞形态观察一致。结论狼疮Ⅰ方具有预防氢化可的松对软骨细胞的损伤作用。

Objective To observe the effects of lupus FangⅠon preventing hydrocortisone damage to chondrocyte. Methods The chondrocyte was isolated from cartilage of 3～4 weeks newly born rabbit by means of collagenase digestion and identified by image analysis, toluidine blue staining, alcian toluidine blue staining, etc. These chondrocyte systems were divided into hydrocortisone groups, traditional Chinese and western medicine groups, traditional Chinese medicine groups and blank groups at random. Different groups of the mixed liquor of lupus FangⅠand hydrocortisone were incubated for 12 hours and 36 hours. Morphologic changes of chondrocyte were observed through inverted microscope. The effect of the mixed liquor of lupus FangⅠand hydrocortisone on the proliferation of chondrocyte was monitored by MTT analysis. Results From observing morphologic change of chondrocyte, chondrocyte were died or major injured in hydrocortisone groups, and the chondrocyte growed well in traditional Chinese and western medicine groups, traditional Chinese medicine groups and blank groups. The results of MTT analysis were showing no difference with chondrocyte amorphous observation. Conclusion Lupus FangⅠhas effect on preventing hydrocortisone damage to chondrocyte.
