环氧化酶-2与泌尿系肿瘤

摘要 环氧化酶(Cyclooxygenase,COX)是一种膜结合蛋白,存在于核膜和微粒体膜,是前列腺素(PGs)生物合成初始步骤中的一个重要限速酶,又称前列腺素内过氧化物合成酶(PGHSs).COX不仅具有脂肪酸环氧化酶的活性还有PGs过氧化的活性,在PGs生物合成过程中,首先是被激活的磷酶水解膜磷脂释放花生四烯酸,接着游离的花生四烯酸由COX催化经过环氧化、过氧化等生成PGH2,PGH2在不同的组织和细胞中代谢生成PGE2、PGI2、血栓烷素A2(TXA2)等产物,几乎所有的PGs和血栓烷素均由PGH2合成.这些产物与各自受体结合,通过调节细胞内环磷酸腺苷(CAMP)、Ca++水平发挥巨大的生物学效应,维持机体的各种生理和病理过程,几乎涉及所有的组织和器官.
