醋酸铅对小鼠精子发生影响的研究 被引量:9

摘要 雄性小鼠分别经饮水染毒0、1000、3162和10000ppm的醋酸铅,染毒后第2周、第7周和第12周每组随机抽取5只雄鼠眼眶取血并分离附睾,进行血铅及精子计数、活动度、形态检查。染毒期内对照组小鼠血铅无明显变化,实验组血铅于染毒第2周即明显增高,并随染毒时间进一步上升,呈良好的剂量效应关系;染毒第2周末,1000和3162ppm组即见精子活动能力和存活率明显下降(P<0.05),其他指标无明显改变;染毒第7周末见3162和10000ppm组精子头部畸形率明显增高(P<0.05);染毒第12周末1000ppm组畸形精子率也见增高,而精子计数下降仅见于10000ppm组长期染毒后。提示醋酸铅可影响小鼠精子发生,其中小鼠精子活力改变是铅生殖毒性最敏感的指标,其次为精子形态改变,而精子计数仅于长期接触高浓度铅方出现改变。 Four groups of male mice, 20 in each group, were administered lead acetate at concentrations of 0,1000,3 162 and 10 000 ppm, respectively. At the end of the second, seventh and twelfth week after administration of the lead, five mice from each group were sampled randomly and sacrificed, Blood was collected and blood and blood lead(PbB) was assayed for contents. Ep.ididymis were isolated and examined for total sperm count, motility and morphology. No remarkable change in PbB waa noticed in control group during the experiment period. PbB in the groups given lead acetate, however, were dose-dependantly elevated significantly. Spermatozoa motility in all the experimental groups was greatly affected after two weeks of Pb administration. Morphological changes in spermatozoa were only found in animals exposed to 1000, 3 162 and 10 000 ppm of lead acetate for twelve weeks. Decrease in sperm count at the end of exposure was also observed in the group exposed to 10 000 ppm of lead acetate, but not in those given 1000 and 3162 ppm.
