摘要
应用荧光法测定血中游离肉毒碱的含量。其原理是通过肉毒碱酰基移换酶(CAT)的酶促反应和N-[P-(2-benzimidazolyl)-phenyl](BIPM)试剂,使之产生强荧光的中间物质,从而测出血中游离肉毒碱的含量。由于采用荧光分光光度法,测定的灵敏度、回收率与重复性均较理想。测定方法简单易行,便于推广。初步临床应用显示糖尿病人的血浆肉毒碱水平明显低于正常人。此外,肉毒碱的测定对于心血管和肾脏等疾病的代谢障碍分析和随访也具有一定价值。
A new fluorometric method for the determination of plasma free carnitine was described. The principal of this method is to measure a strong fluorescence from a intermediate substance by the reaction of carnitine acetyltransferase and BIPM indicating the free carnitine level. The fluorospetrophotometric assay is sensitive and reproducible. Its recovery rate is ideal and the procedures are easy and simple. The preliminary results suggest that the plasma carnitine level in diabetic patients was obviously lowered than in normal subjects. The measurement of carnitine was also valuable infollow up and assassment of metabolic disorder in patients with cardiovascular disease and nephropathy act.
