免疫组织化学法研究大脑皮质创伤后室管膜细胞及其下层的细胞变化

摘要 65只成年雄性大鼠,切除大脑皮质顶叶正中部分(4×3×1mm),术后第4d取材。其中10只大鼠用胶质原纤维酸性蛋白抗体,30只用烯醇酶抗体,25只用神经丝抗体,应用免疫组织化学方法显示室管膜细胞和室管膜下层的细胞。位于损伤区邻近的室管膜细胞呈胶质原纤维酸性蛋白免疫染色阳性,在室管膜细胞间可见有伸展细胞,也呈胶质原纤维酸性蛋白阳性,并具有特殊的形态。与损伤区邻近的室管膜下层,细胞出现增生分裂现象,个别细胞的胞体较大,直径约20~24μm,这些大细胞经用烯醇酶抗体和神经丝抗体免疫染色均呈阴性,胶质原纤维酸性蛋白免疫染色则出现强阳性。这些细胞可能是分化较低的星形胶质细胞。 Sixty five adult male rats were injured by partial decortication (4×3×1mm wound size), four days later all animals were killed. The ependymal cells and subependymal layer were Observed with immunostaining methods including 10 rats stained for glial fibrillary acidic protein (GFAP), 30 rats for neuron-specific enolase (NSE) and 25 rats for neurofilaments (NF). Results: The ependymal cells of lateral ventricle near the damaged area were stained with GFAP, and they were GFAP positive. In some area tanycytes developed which were of special morphology. Proliferation and mitosis were shown in subependymal layer of lateral ventricle near the damaged area. The diameters of the larger cells were about2024μm. The larger cells were NSE and NF negative, but staining for GFAP was positive. These cells might be astrocytes in poor differentiatio
