摘要
<正> 尿激酶(Urokinase)简称UK[E.C.3.4.21.31]是一种纤维蛋白溶酶原激活剂,在临床上可用于治疗血栓,存在于正常人的尿液中。按国内目前普遍使用的尿激酶精制工艺收率低,仅为40%左右,产品比活最高仅达4—5×10~4IU/mg蛋白,而且工艺流程长、设备投资大。
Hybridoma technique was used to establish some cell lines of bybridoma secreting monoclonal antibodies against urokinase. One of the monoclonal antibodies (1B4) was purified and coupled to Sepharose 4B,which served asmatrix to purified crud urokinase with original specific activity 1000IU/mg Protein. After one step purification the specific activity of urokinase increased to over 8×10~4IU/mg. protein, reaching highly purified degree. Its total recovery was over 60%.
