摘要
钙调蛋白是生物细胞内一种重要的调控蛋白,通过其与靶酶的相互作用,控制细胞正常的生长和发育。我们以湖北光敏感核不育水稻cDNA第一条链为模板,参考大麦钙调蛋白基因序列合成5'端和3'端引物,利用多聚酶链式反应合成了完整的水稻钙调蛋白cDNA,克隆并测定其序列。结果表明,光敏核不育水稻的钙调蛋白基因由450个核苷酸组成,共码148个氨基酸。
Calmodulin is a calcium binding protein that modulate the activity of a diverse group of proteins including some protein kinases, adenylate cyclases and ATPase. Here we report the cloning and sequencing of the full-length cDNA encoding calmodulin mRNA from rice (Oryzae sativa). The first strand cDNA was synthesized using rice leaf mRNA in the present of 26 and 24 nucleotides as primers according to barley cam sequence. The calmodulin gene was amplified through PCR before it is actually clond. We found that calmodulin gene consists of 450 bp and the protein as predicted from DNA sequence, consists of 148 amino acids and differs from that of barley and alfalfa in only 1 position Seperately. The nucleotide homology between rice and barley is 90%, between rice and alfalfa is 88%. Calmodulin is a very conservative regulator protein.
