摘要 冰冻人血浆37℃融化后,经钡盐吸附沉淀、硫酸铵分步盐析、DEAE-Sephadex A-50柱层析、制备性PAGE和肝素亲和层析分离,得到蛋白C。经鉴定,所得蛋白C分子量约为62kD,pI为4.69,PAGE分析高度均一,KPTT法证实其延长凝血时间,与有关文献报道相符。 Protein C was isolated by means of barium salt adsorption, ammonium sulfate fractionation, DEAE-Sephadex A-50 column chromatography, preparative PAGE and haperin affinitive chromatography from frozen human plasma, which was thawed at 37℃. The molecular weight and pI of the protein C are 62kD and 4.69, respectively. The purified protein C is shown to be highly homogeneous by analysis of PAGE and to be active by analysis of KPTT.
