摘要
本文用低浓度氯化钠与肝脏一起匀浆,75℃加热,硫酸铵分级沉淀,在Cu^(2+)存在下透析,sephadex G-75柱层析等方法,从寒鸦肝脏中纯化出铜锌超氧化物歧化酶。对其理化性质鉴定表明,用此法纯化的SOD为均一性纯酶,比活性为4734U/mg pr,分子量32.6kD,紫外吸收峰在258.6nm。理化性质与文献报道的不同来源的同类酶基本相同。
In this paper copper-zinc superoxide dismutase was purified from crow liver by short time homogenate with sodium chloride at low concentration, heat treat ment at 75℃, dialysis in the presence of Cu^(2+), following precipitation with aminonium sulfate and sephadex G-75 column chromatography. Through.the investigation of physiocheraical properties, the purified SOD by this method is homogvneonsly pure. Its specific activity is 4734U/mg determined by means of pyrogallol autoxidation. Its molecular weight is about 32.6 kD. A maximum absorption of the enzyme is found at 258.6nm.These properties of the crow liver CuZn-SOD are very similar to those already reported about the Cu·Zn-SOD from other sources.
