摘要
本实验通过逆转录病毒载体pMV7将大鼠蛋白激酶c(Protein Kinase C,PKC)βⅠ亚类cDNA全长片段导入NRK细胞中,建立了一个过表达PKC-βⅠ亚类的NRK细胞模型,并对此细胞模型在佛波脂TPA进一步诱导激活PKC状态下的生长情况及与c-jun基因表达的相关性进行了初步观察。
We utilized a retroviral expression vector to construct a series of NRK cell lines that overexpress a full length cDNA encoding rat protein kinase C β I (PKC β I). These cell lines contain 3—4 fold greater PKC activity than parental cells or control cells that carry an integrated vector lacking the cDNA insert. When expression of PKC was further activated by TPA, the growth conditions and the level of c-jun gene expression in these cell lines were also observed.
