摘要
3~H-TdR放射性转化细胞(TC3H/10)经1×10^(-5)mol/L Foskolin处理24 h后,TGFα、c-myc、c-K-ras基因的mRNA表达下降,TGFβ、c-fos基因表达无明显变化。非转化细胞(NC3H/10)经相同条件处理,TGFα、TGFβ、c-myc及c-K-ras基因表达无显著改变。提示:Forskolin介导的转化细胞的生长抑制作用与TGFa、cmyc、c-K-ras基因的转录表达下降有关。
The effects of FSK on the expression of TGFα/β genes and some oncogenes in ~3H-TdR transformed C_3H/10 cells were investigated. The results showed: 1. FSK(1×10^(-5)mol/L, 24 hours) inhibited the mRNA expression of TGF_α c-myc,and c-K-ras, and failed to reduce TGF_β and c-fos mRNA levels in TC_3H/10, cells. The expression of TCF_α, TCF_β, c-myc,and c-K-ras in NC_3H/10 cells treated by FSK did not change siginificantly. 2. The soft agar cloning formation and ~3HTdR incorporation of TC_3H/10 cells were inhibited by FSK. These results suggested that inhibition of TGF_α, c-myc and c-K-ras gene expression by FSK is responsible for the decreased proliferation of the TC_3H/10 cells.
