摘要
用荧光光谱方法研究了江浙蝮蛇毒中性磷脂酶A_2(NPLA_2).研究结果表明NPLA_2分子中确实含有一个色氨酸残基.且位于NPLA_2分子表面;我们还发现荧光探针bis-ANS在NPLA_2分子上有一结合区,其解离平衡常数为11.6μmol/L ;利用结合了的bis-ANS与NPLA_2分子中色氨酸残基之间的能量传递计算出两者之间的距离为17.7(?).
The neutral phospholipase A2 from the venom of Agkistrodon halys Pallas (NPLA2) was studied by fluorescence spectroscopy in this paper. The results showed that one tryptophan residue did exist in NPLA2 and located near the surface of the NPLA2 molecule. We also observed that NPLA2 contained a single binding site for the fluorescence probe-1.1'-bi -(4-anilino) naphthalene-5.5'-disulfonic acid (bis-ANS) and its dissociation constant was 11.6 μmolL. The radiationless energy transfer from the tryptophan residue in NPLA2 to bound bis-ANS was used to estimate the distance between these two groups, which was 17.7A.
