摘要
目的 研究骨髓基质细胞体外培养的成骨能力 ,为细胞移植修复骨缺损提供一条新途径。方法 取新西兰大白兔股骨大转子处骨髓 ,进行体外培养 ,纯化 ,10~ 14天后进行传代。培养后的细胞经倒置显微镜、透射电镜、钙染色、碱性磷酸酶染色等方法进行观察。结果 培养纯化后的细胞呈梭形、多角形 ,电镜显示具有分泌功能旺盛的结构 ,碱性磷酸酶染色强阳性。培养 3~ 4周后 ,能形成钙结节。细胞在体外能多次传代 ,维持成骨表型。结论 骨髓基质细胞在体外培养纯化后 ,具有与成骨细胞相似的形态结构和生物学特性 。
Objective: To study the osteogenic potential of bone marrow stromal cells (BMSC) and find a new model of repairing segmental bone defects. Methods: Bone marrow was obtained from the trochanteric region of femur of rabbits and cultured in vitro. The hematopoietic component was discarded, and then pure BMSC was obtained. The cells were examined by phase-contrast microscope, transmission electron microscope, salvaria stain, alkaline phosphatase assay(ALP). Results: Pure BMSC lined up in one layer and formed nodules of calcification in vitro. The secretion and ALP activity were more active. The cultured cells maintained osteogenic phenotype for a long time. Conclusion: The morphological and biological characteristics of the cultured stromal cells are similiar to osteoblasts and can synthesize bone tissue in vitro.
