急性硒中毒后小鼠肝细胞线粒体损伤的时间形态学研究 被引量:3

摘要 对急性二氧化硒中毒所致肝细胞线粒体损伤进行时间形态学定量研究 ,以发现其敏感时间点 ,为临床合理地使用硒制剂提供基础资料。雄性BALB/C小鼠 60只 ,随机分为正常对照组与染毒组 ,每组 30只。每组再随机分为 6个时间组 ,每隔 4小时各一组 ,每组 5只。染毒组小鼠以口服LD50量腹腔注射二氧化硒 (SeO2 )。两组均在 1小时后处死。应用体视学方法对肝细胞线粒体的体密度 (Vv)、比表面 (δ)、表面积密度 (Sv)、平均体积 (V)和数密度 (Nv)进行测算。硒中毒后小鼠肝细胞线粒体与对照组相比 ,δ除 0 4 :0 0组外 ,其他时间组则明显减小 (P <0 .0 5) ,最低值出现在 12 :0 0 ;V除 0 8:0 0组外 ,其他时间组普遍增大 ,其中 12 :0 0组明显增大 (P <0 .0 5)。Nv 在 12 :0 0、16:0 0和 2 0 :0 0组与对照组相比明显减少 (P <0 .0 5) ,其他时间组与对照组相比则未见显著性差异 (P <0 .0 5) ;而体密度和表面积积密度则无明显改变。硒中毒可直接导致肝细胞线粒体的损伤 ,表现为线粒体体积增大 ,比表面及数量减少 ,从而导致其功能降低。硒中毒时其线粒体损伤变化表现为夜间较轻 ,白天较重。 0 4 :0 0h可能为肝细胞线粒体的不敏感时间点 ;而 12 :0 0h可能是其硒中毒的最敏感时间点。 The aim of present study of the relationship between hepatocyte mitochondria morphological variety time on mice following SeO 2—Poisoning was to determine the sensitive time distribution spot for rational usage selenium in clinic. Sixty male BALB/C mice were randomly divided into control group and poisoned group(30 mice each),which were randomly subdivided into six groups(5 mice each)for four hours every other within 24 hours. The group of mice which were given Selenium Dioxide(oral LD50)by intraperitoneal injection is the poisoned group. The mice were executed after one hour. The volume density(Vv),speciflc surface(δ),surface density(Sv),mean volume(V)and numerical density(Nv)of mitochondria were analysed with Stereology. The δ, V and Nv of mitochondria were significant different between control group and Selenium poisoning group. The δ of mitochondria in each poisoned group was significant decreased(P<0.05),except for only one group(4:00 am group,P>0.05),and the smallest δ was at 12:00am. The V of mitochondria was generally increased and the highest value was at 12:00 (P<0.05) after selenium poisoning. The Nv of mitochondria was significantly decreased at 12:00,16:00 and 20:00 when compared with that of normal group(P<0.05). But the Vv and Sv of mitochondria were not significantly changed. Selenium poisoning can directly lead to hepatocyte mitochondria damage. Mitochondria morphological variety after acute selenium poisoning showed their increased mean volume, decreased specific surface and numerical density,which lead to the function of hepatocyte mitochondria weakened. There was obviously day and night rhythms of the mitochondria damage after selenium poisoning,showing the damage was weaker at night than that of at day. It is inferred that the 04:00 am is the non-sensitive time spot and the l2:00 pm is the most sensitive time spot of selenium poisoning. It provides a new clue for recognition and prevention Selenium poisoning.
