急性胰腺炎大鼠胰腺组织中转化生长因子及Ⅰ型胶原变化的研究

Changes of transforming growth factor and collagen type I in pancreatic tissue after acute pancreatitis in rats

目的 :研究转化生长因子 (TGF)对急性胰腺炎发生后胰腺组织修复的影响。方法 :雄性Wistar大鼠 96只 ,实验组和对照组各 48只 ,腹腔一次性注射L 精氨酸制成胰腺炎模型 ,分别于术后 4、12、2 4、48、72、96h各处死 8只取标本 ,进行病理学检查、血淀粉酶的测定及胰腺组织TGFβ1mRNA及Ⅰ型胶原mRNA的检测 (Northern分子杂交方法 )。结果 :腹腔注射L 精氨酸后经病理学及血淀粉酶检测证实产生了急性水肿性胰腺炎的病理损害 ,但同时胰腺组织内TGFβ1mRNA水平升高 ,于 12h时达到较高水平 ,持续至 72h时开始下降 ;这一时段内Ⅰ型胶原mRNA水平也显著升高 ,并与TGFβ1mRNA的变化呈密切相关性 (r=0 72 8,P <0 0 5 )。结论 :在急性胰腺炎胰腺组织病理损害的同时存在着胰腺自身修复的机制 。

To study the effect of transforming growth factor(TGF)on pancreatic repair after acute pancreatitis.Methods: Pancreatitis were induced by intraperitoneum injection of L Arginin in 48 Wistar rats,8 rats were killed at the point of 4,12,24,48,72 and 96h,and samples were harvested. Pathologic observation, detection of serum amylase and TGF β 1 mRNA, collagen type Ⅰ mRNA(northern blot analysis)were proformed.Results: Animal models acute pancreatitis were proved by pathologic observation and serum amylase detection. The level of TGF β 1 mRNA increased in pancreatic tissue,reaching the peak at 12h, then bean to decline at the point of 72h; at the same time, the level of collagen typy Ⅰ mRNA increased too, and had intimate correlation with TGF β 1 mRNA(r=0 728,P<0 05).Conclusion: There exists pancreatic self repair mechanism in acute pancreatitis and TGF coooagen type Ⅰ play important role in it.
