摘要 目的 :探讨凋亡相关基因Fas/FasL在甲状腺癌组织中的表达与细胞凋亡的关系。方法 :采用流式细胞技术检测 43例乳头状甲状腺癌癌细胞凋亡率及相关基因Fas及FasL表达。同时检测Fas及FasL在甲状腺癌组织肿瘤浸润淋巴细胞 (TIL)中的表达。结果 :43例乳头状甲状腺癌细胞中Fas低表达者 19例 ,细胞凋亡率为 3 71% ;高表达者 2 4例 ,细胞凋亡率为 7 2 6 % ,高表达组癌细胞凋亡率明显高于低表达组 (P <0 0 5 ) ,癌细胞中FasL表达明显高于甲状腺腺瘤组织 (P <0 0 1) ,乳头状甲状腺癌组织TIL表达Fas及FasL的水平明显高于甲状腺腺瘤组织 (P <0 0 1)。结论 :Fas系统参与乳头状甲状腺癌组织中细胞凋亡的调节 ,Fas/FasL表达异常使甲状腺癌癌细胞逃避免疫监视 ,诱导Fas敏感的TIL凋亡 ,有助于癌细胞发生浸润及转移。 Objective: To investigate the expressions of Fas, FasL in papillary adenocarcinoma of thyroid gland tissues and their tumor infiltrating lymphocytes(TIL), and its relationship with apoptosis and Fas, FasL in thyroid tumors.Methods: We detected the apoptotic rate and the expression of apoptotic related gene Fas in 43 cases of papliiary adenocarcinoma of thyroid gland with flow cytometry. The expression Fas and FasL were measured in 43 cases of papillary adenocarcinoma of thyroid gland, 28 cases of thyroid adenoma,43 paplillary adenocarcinoma of thyroid gland TIL,17 thyroid adenoma TIL using flow cytometry.Results: 19 cases in papillary adenocarcinoma showed low expression of Fas protein with an average apoptotic rate of 3.71% and 24 cases had high expression of Fas with an apoptotic rate of 7 26%(P<0 05). FasL was higher expression in paplillary adenoma tissues than that in thyroid adenoma(P<0 01). There were abundant TIL in papillary adenocarcinoma tissues, the expression of Fas and FasL in papillary adenocarcinoma TIL were higher than that in thyroid adenoma TIL(P<0 01).Conclusion: Fas and FasL take part in the occurrene and progression of papillary adenocarcinoma. Tumor cells may escape from immune surveillance by highly expressing FasL and inducing TIL apoptosis and perform infiltration and metastasis.
