摘要
目的 :探讨精神应激 (天敌应激 )对小鼠腹腔巨噬细胞产生细胞因子的影响 ,进一步完善神经免疫内分泌网络中下丘脑 垂体 肾上腺 单核 巨噬细胞 (HPA Mo MΦ)调节环路的知识。方法 :将小鼠分为正常对照组、急性应激组和慢性应激组。应激组暴露在猫境中。急性应激 [4 5min (次·d) ]和慢性应激 [4 5min (次·d) ,连续刺激 1 4d]后 ,快速断头杀鼠 ,无菌制备小鼠腹腔巨噬细胞 ,经 2 4h培养后用酶联免疫吸附测定法测定腹腔巨噬细胞培养上清液中IL 1 β、IL 6蛋白水平。结果 :慢性天敌应激后培养上清液中IL 1 β、IL 6水平较正常对照组无明显变化 (P >0 0 5 ) ;急性应激后 ,水平明显降低 ,与正常对照组及慢性应激组比有统计学意义 (P <0 0 5 )。结论 :天敌应激可影响巨噬细胞功能以及HPA Mo MΦ调节环路活性。
Objective:To explore the effect of psychological stress(predatory stress)on cytokines produced by peritoneal macrophage.Methods:Mice were divided into groups of normal control,acute and chronic predatory stress.Stressed mice received either a single 45 minutes cat exposure(acute predatory stress group)or daily 45 minutes exposure to the cat for consecutive 14 days(chronic predatory stress group).Subsequently,all mice were sacrificed by rapid decapitation and peritoneal macrophages were prepared and then cultured for 24 hours. The supernatants of cultured peritoneal macrophage were collected and then used for detection of IL-1β and IL-6 levels by enzyme-linked immunnsorbent assay.Results: For IL-1β and IL-6 levels in the supernatants, there was no significant alteration observed in chronic stress group, compared with normal control group(P>0.05). Whereas, IL-1β and IL-6 levels were notable decreased after acute predatory stress, revealing a significant difference relate to the groups of normal control and chronic stress(P<0.05). Conclusion: Predatory stress can influence the function of macrophage and the activity of HPA-Mo/MΦ circuit in Balb/c mice.
