异丙酚对大鼠肠源性肺损伤Fas、Caspase-3的影响

Effect of Propofol on the expression of Fas and Caspase-3 in Acute Lung Injury Following Small Intestine Ischemic Reperfusion in Rats

摘要 目的 研究异丙酚对急性肺损伤时Fas、Caspase - 3的影响。方法 健康SD大鼠 36只随机分 3组 ,空白对照组 (S组 )、缺血再灌注组 (I/R组 )、异丙酚组 (P组 ) ,每组 12只。采用小肠缺血再灌注复制急性肺损伤模型。光镜观察组织病理改变 ;免疫组织化学检测Fas和Caspase - 3的蛋白表达 ,并测量光密度值 (OD值 ) ;原位末端标记法(TUNEL)检测凋亡的肺细胞并计算凋亡指数。结果 I/R组Fas和Caspase - 3OD值明显高于S组和P组 (P <0 0 1) ,且P组Caspase- 3OD值高于S组 (P <0 .0 5 ) ;I/R组细胞凋亡指数明显高于S组和P组 (P <0 0 1) ,而P组高于S组 (P <0 0 5 )。结论 异丙酚能降低Fas和Caspase - 3蛋白的表达 ,抑制细胞凋亡 ,保护急性肺损伤。 Objective To study the effect of propofol on the expression of Fas,Caspase-3 in acute lung injury following small intestinal ischemic reperfusion.Methods 36 SD rats were randomized into three groups each consisting of 12 rats:sham group(S group),Ischemic Reperfusion group(I/R group),Propofol group(P group).Acute lung injury models were made by ligated the small intestinal superior menseneric artery.The expression of Fas and caspase-3 were detected using immuhischemistry.Apoptotic lung cells were measured by TUNEL method,and calculated the apoptotic index.Results The average OD value of Fas and Caspase-3 were significantly higher in I/R group than in S and P group(P<0.01),and the average OD value of Caspase-3 was higher in P group than in S group (P<0.05).The apoptotic index in I/R group was significantly higher than that in other two groups(P<0.01),and that in P group was higher than S group (P<0.05).Conclusions Propofol can protect lung injury associating with inhibition of the expression of Fas , Caspase-3.
