摘要
在无反应发生条件下进行固定化细胞内组分有效扩散系数测定时,常常要使细胞失去活性。本文通过显微形态观察及重量变化测定,分析了常用的几种失活细胞的方法对细胞本身的影响。测定了由各种不同方式得到的无活性固定化细胞内葡萄糖与乙醇的有效扩散系数,并与有活性固定化细胞内组分的有效扩散系数进行了比较。提出了一种便于使用,既可保护底物不被消耗,又可以测得更接近实际值的有效扩散系数的无反应发生条件:苯甲酸处理法。
There exist different methods of inactivating cells which give a non-reaction-occurring condition under which the effective diffusivity of a substrate in the immobilized cell matrix is often measured. Based on micrographic observation of size and shape of the cell and determination of weight change of cells,the effect of the various inactivating methods on the cell itself was examined. The effective dif-fusivities of glucose and ethanol in the inactivated yeast cell-occupied polyacrylamide gels obtained from the various methods were determined and compared with the diffusivity in the active immobilized cell. So the method called as 'benzoic acid-treating method'has been proposed which is convenient to work,provides a non-substrate-consuming condition and gives a more reasonable value of diffusivity than other methods .
