Synthesis of Linoleic Acid Hydroperoxide Catalyzed by Soybean Lipoxygenase

摘要 研究了水相体系中大豆脂肪氧合酶酶促合成亚油酸(LA)氢过氧化物的反应,最佳反应条件为底物质量浓度4 11g/L,pH=9,5℃,反应时间120min,亚油酸氢过氧化物产率80%。当m(LA)∶m(surfactant)=1∶1时,Tween系列、CTAB、AOT、AEO4、OAPCG、S8390均能加速反应,其中AEO4使反应时间缩短至30min,产率提高12%。一些盐可以明显地加快反应进程,其中柠檬酸钠使反应时间缩短至15min。外源Fe3+在反应体系中的浓度为0 5μmol/L或外源Fe2+为5nmol/L时反应速度显著提高,消除了滞后期。 Synthesis of linoleic acid hydroperoxide(HPOD) with linoleic acid(LA) catalyzed by soybean lipoxygenase(LOX) in aqueous phase was studied.Thus,4.11 g/L LA in buffer solution of pH=9 reacted at 5 ℃ for 120 min to give HPOD in 80% yield.Some external factors can promote the LOX catalysis.Surfactants including Tweens,CTAB,AOT,AEO_4,OAPCG and S8390 in the ratio m(LA)∶m(surfactant)=1∶1 accelerated the reaction markedly.Addition of AEO_4 shortened the reaction time to 30 min with 12% increase of yield.Some salts can promote LOX catalysis,among which sodium citrate(5 g/L) shortened the total reaction time to 15 min.Exogenous Fe^(3+) of 0.5 μmol /L or exogenous Fe^(2+)of 5 nmol / L eliminated the lag phase and accelerated the catalytic reaction.
