摘要
目的 为了分离孕妇外周血中的有核红细胞用于非损伤性产前诊断。方法 采用了一种新的亲合素法 ,用红细胞表面半乳糖基特异的凝集素 (大豆凝集素 )来选择富集孕妇外周血中的有核红细胞。结果 从 11份 2 .5ml孕 14 + ~ 15 +w的孕妇外周血中都分离到了有核红细胞 ,平均每份血样得到有核红细胞 2 .8个。结论 本方法实验成本低 ,操作简单 ,对于非损伤性产前诊断的研究和推广都具有重要的参考价值。
Objective: To isolate nucleated red blood cells (NRBCs) from maternal blood. Methods: A new technique using galactose-specific lectin was adopted, which based on galactose expression on the surface of NRBC. Results: NRBCs were successfully isolated from 11 blood samples of pregnant women at 14 +~15 + gestational weeks. The average of 2.8 NRBCs were obtained from 2.5ml blood sample. Conclusion: A simple and sensitive method was presented for isolation of NRBCs in maternal blood, which may be helpful for clinical application of noninvasive prenatal diagnosis.
