摘要
目的 探讨年轻细胞被诱导衰老后对凋亡的敏感性与年轻细胞的差异 ,并分析可能存在的机制。方法 以人胚肺二倍体成纤维细胞 (2BS)为研究对象 ,以H2 O2 诱导 2BS细胞早衰后 ,再以H2 O2 分别诱导早衰 2BS细胞和年轻细胞凋亡 ,DNAladder分析和苔盼蓝排除法检测细胞死亡率。结果 早衰细胞较年轻细胞不易被H2 O2 诱导凋亡。该因素是多方面的 ,不限于caspase - 3的作用。 结论 早衰细胞与衰老细胞一样 。
Objective To explore the difference susceptility to apoptosis of young cells before and after being induced senilism,and analysis the possible mechanism.Methods Human diploid fibroblasts are investigated in this study.After H 2O 2 was used to induce senilism in human diploid fibroblasts(strain 2BS),then to induce apoptosis in senilism cells and young ones.Apoptosis was detected by DNA ladder analysis and taipan blue excluding.Results The results showed that senilism cells are more resistant to apoptosis by H 2O 2 than young ones.The reasons is in many ways,it is not confined to function of caspase-3.Conclusion Senilism cells are sililar to serescent cells,reduce the susceptility to apoptosis.
