The hypocotyl protoplasts of Lyciumbarbarum L.CV.Ningji No.1 were isola-ted in an enzyme mixture solution contain-ing 1% cellulase and 1% pectinase.Theprotoplasts were cultured in KM 8P liquidmedium containing 0.3mg/L 2,4-D and0.3mg/L BA,The first division of rege-nerated cells occurred 3 days after culture.The small cell clumps could be observed bynaked eyes 20 days after culture.40 daysafter culture,microcalli of 1-2mm in sizewere transferred to MS solid differentiationmedium containing 3% sugar,0.1-0.4mg/LBA and 0.05-0.1mg/L NAA.About 30days after culture on MS solid medium,the calli with shoots were transferred to1/2 MS medium containing 0.1mg/L BAand 0.2mg/L IBA,from which 80% ofthem differentiated roots and regeneratedwhole plantlets.
