摘要
本文利用无血清培养的末成年大鼠前列腺上皮细胞模型研究了催乳素(PRL)和性激素对前列腺细胞分泌前列腺特异的酸性磷酸酶(PAP)功能的作用,并对其作用途径进行了探讨。结果表明:PRL 显著促进前列腺细胞PAP的合成和分泌,但与双氢睾酮(DHT)或雌二醇(E_2)无协同作用,单独的DHT 或E_2对PAP 的合成和分泌也无显著作用。在PRL 的作用过程中,腺苷酸环化酶未活化;PRL 和前列腺素E_2或F_(2α)有协同作用,消炎痛能阻断PRL 的作用。综合实验结果,本实验证明了PRL 对前列腺细胞的分泌功能有直接的刺激作用,在PRL 的作用过程中,可能有前列腺素的参与。
The present study revealed the effectof prolactin(PRL)and sex steroid hormo-nes on production of prostatic acid phos-phatase(PAP)of the immature rat prosta-tic epithelial cells in serum-free medium.The results showed that PRL significantlystimulated synthesis and secretion of PAP,but failed to synergize with dihydrotestos-terone or estradiol.Dihydrotestosterone orestradiol alone did not promote PAP pro-duction.Studies on the mechanism of PRLaction indicated that adenyl cyclase wasnot activated in the action of PRL:Syner-gistic effect between PRL and prostaglan-din E_2 and F_(2α)was revealed in the stimu-lation of PAP production,and the effectof PRL could be blocked by indomethacin(a prostaglandin synthesis inhibitor).Inconclusion,PRL directly stimulated PAPproduction of the cultured rat prostaticepithelial cells in serum-free medium andprostaglandin was involved in PRL action.
