真菌细胞破壁方法的研究

Study on the Methods of Breaking Wall of Fungus Cell

用4种处理对真菌细胞进行破壁,使其释放出细胞内物质.处理后的菌液用血细胞计数板计数,得到每毫升菌液中的孢子数和菌体碎片数,观察比较机械破碎的效果.以可溶性蛋白为参照,通过蛋白质的SDS-PAGE分析,比较4种处理方法使细胞破壁后释放蛋白质效果的差异.结果表明,方法2的破碎效果和细胞可溶性蛋白质释放效果都较好,是一种首选真菌细胞破壁的方法.

Four kinds of method for breaking fungus cell walls were used to release proteins in the fungi. The spore and fragment numbers in the germ solution dealed with the above methods were determined with blood corpuscle counting meter. With releasing soluble protein as example, the differences of releasing proteins by the four methods of breaking cell walls of fungus were analyzed with SDS-PAGE. The results showed that the second method, i.e. breaking wall with ultrasonic and adding silica sand, appeared well effect and it was the first selective means for breaking cell wall of the fungi.
