血浆中右旋酮洛芬的HPLC测定

Determination of dexketoprofen by HPLC

目的采用高效液相色谱法测定家兔血浆中右旋酮洛芬浓度。方法用乙醚提取血浆样品中右旋酮洛芬,在C18柱上,以甲醇50mmoL·L-1磷酸二氢钠(30∶70)为流动相进行分离,于260nm检测。结果线性范围为005～25μg·mL-1,回收率为9992%～10054%,日内RSD为056%～294%,日间RSD为06%～289%。结论本法简便、准确,重现性好,用于测定6只家兔口服右旋酮洛芬β环糊精缓释微球血药浓度取得良好结果。

OBJECTIVE: To develop a HPLC assay for determining dexketoprofen in the rabbits plasma. METHODS: Dexketoprofen was extracted with ether. C18 column was used and the mobile phase consisted of methyl alcohol - 50 mmoL&middotL-1NaH2PO4(30:70). The detection wavelength was at 260 nm. RESULTS: The range of 0.05 ∼ 25 μg&middotmL-1 was obtained with good linearity. The recovery was 99.92% ∼ 100.54%. The interday validation and the intraday validation was 0.56% ∼ 2.94% and 0.6% ∼ 2.89%, respectively. CONCLUSION: The HPLC assay was simple, convenient, accurate, and reproducible. It had been used to determine the dexketoprofen concentration in rabbit plasma after administering dexketoprofen-β-cyclodextrin inclusion microspheres orally.
