摘要
目的 :双氢青蒿素作用于人喉癌细胞 (HeP 2 ) ,观察药物对其生长抑制作用 .方法 :采用细胞计数法、MTT法、软琼脂克隆形成试验等观察、测定双氢青蒿素对喉癌细胞 (HeP 2 )的细胞增殖率、细胞群体倍增时间、双氢青蒿素对HeP 2细胞生长抑制作用的剂量 -效应曲线、时间 -效应曲线以及对该细胞克隆形成率的影响 .结果 :①细胞生长曲线显示该细胞增殖活跃 ,生长稳定 ,细胞群体倍增时间为 2 4 7h ;②双氢青蒿素对Hep 2细胞有明显的生长抑制作用 ,其作用特点呈浓度依赖性 ,其IC50 值为 4 4 6 6 84 μg/L .时间 -效应曲线显示 :当作用时间在 2 4~ 12 0h时 ,呈剂量依赖性关系及时间依赖性关系 .③双氢青蒿素药物浓度分别为 32 0、10 0 0、32 0 0、10 0 0 0μg/L时 ,克隆形成率分别为 1 15 0、0 9、0 6和 0 5 (% ) .结论 :双氢青蒿素对HeP 2细胞增殖及其生长有显著的抑制作用 ,随着药物作用时间的延长 ,对于细胞的生长抑制作用逐渐增强 ,表明双氢青蒿素在喉癌的化学治疗中对肿瘤细胞有直接的细胞毒作用 .
AIM: To observe the growth inhibitory effects of pair of hydrogen artemisinin on HeP-2 cells of throat cancer. METHODS: Cell proliferation was studied by MTT assay and soft agar cloning test. Different concentrations of pair of hydrogen artemisinin were applied, and IC 50 and the cloning ratio of the drug against the cells were calculated. RESULTS: Pair of hydrogen artemisinin inhibited the proliferation of Hep-2 in a time and dose dependent manner. The IC 50 of the drug against the cells was 4466.84 μg/L, and the cloning ratio (%) of the cells exposed to the drug at 320, 1000, 3200 and 10000 μg/L was 1.150%, 0.9%, 0.6%and 0.5% respectively, while that of the control was 1.2. CONCLUSION: Pair of hydrogen artemisinin inhibits the growth of Hep-2 cells.
