摘要
目的 :研究IGF -I对培养的新生小鼠细胞的促肥大作用及信号转导途径 ,方法 :测定了IGF -I作用下 [4 5-3 H]亮氨酸在心肌细胞的参入及 genistoin ,PD980 5 9,wortmannin和chelerythrinechloride的阻断作用 ,结果 :IGF -I明显地促进增加心肌细胞的 [4 5 -3 H]亮氨酸的掺入作用 ,而且这种作用可以被 genisteinPD980 5 9和chelerythrinechloride阻断 。
To study the effects of IGF-I on hypertrophy in primary cultured neonatal rat cardiomyocytes,and to determine the pathway of signal transduction of IGF-I in stimulating hypertrophy of cardiomyocytes.[4 5- 3H] Leu incorporation assay,and the photo of phase contrast microscopy inspection were done.Genistein,PD98059 wortmannin,and chelerythrine chloride in cardiomyocytes were observed.Results showed that IGF-I can increase [4 5- 3H] Leu incorporation significantly in cardiomyocytes,and that effects were inhibited by genistein,PD98059 and chelerythrine chloride,but not by wortmannin.
