IFN-γ、IL-12对B7-1转染肝癌细胞诱导T淋巴细胞活化的影响 被引量:3

摘要 目的 探讨B7-1基因转染肝癌细胞与IFN-γ、IL-12联合在体外诱导T细胞活化的能力。方法酶联免疫吸附反应(ELISA)检测联合诱导瘤苗刺激淋巴细胞IFN-γ、IL-12蛋白合成及细胞表面CD4、CD8、CD25的表达。MTT法检测瘤苗对淋巴细胞增殖作用的影响。结果 HepG2/B7-1细胞诱导淋巴细胞表达IL-2、IFN-γ,其蛋白质含分别为875 pg/ml和1 246 pg/ml;而联合IL-12、IFN-γ后能进一步诱导IL-2、IFN-γ的蛋白质合成,较各自无细胞因子组明显提高(P<0.05)。细胞表面CD4、CD8、CD25的表达显著增加。两者联合促进了淋巴细胞的增殖,在E/T=10:1时,较HepG2组明显增大。结论IL-12、IFN-γ增强经B7-1基因转染的肝癌细胞(HepG2/B7-1)诱导淋巴细胞产生IL-2、IFN-γ的能力;IL-12、IFN-γ可能通过调节B7-1分子的生物学活性影响其对T细胞的活化。 Objective To evaluate the capability of B7-1 costimulatory molecule alone or combined with IFN-γ/IL-12 in inducing T lymphocytes activation in vitro. Methods ELISA techniques were used to assay the production of IFN-γ and IL-2 and expression of CD4, CD8 > CD25 in lymph-cells. Lymphocyte proliferation was evaluated by MTT assay. Results The combination of HepG2/B7-1 with IFN-γ/IL-12 promoted the proliferation of lymph-cells, which was obviously accereted when E/T = 10 : 1 compared with HepG2 alone. Conclusions IL-12 and IFN-γ may enhance the activation the lymphocyte induced by B7-1 gene transfered hepatccarcinoma cells, which may be related to an increase of proliferation of lymph-cells.
